The heat is on: How does the endometrium mediate the health impacts of climate change associated heat stress?  (141562)

One of the most pressing issues facing the planet today is climate change. Within the climate change context, we know that heat stress (HS) impacts human health. However, the impact of heat stress during the peri-conception period, and more specifically during the peri-implantation on the mother during pregnancy and the potential life-course health implications for the health of the child remains less well understood. This is a difficult period of pregnancy to study in humans, with follow up studies on child health difficult. We are using a pig model of HS in vivo to test how peri-implantation impacts the health and adaptation of the maternal environment to HS during the time when body patterns are being established in the embryo. We have mapped these HS exposed changes in the endometrium using single nuclei-multiomic analyses. We have generated endometrial organoids from HS and control individuals and tested how they respond to key molecules for early pregnancy success. Along with this we have established novel in vitro models of varying complexity including extracellular scaffolds, and endometrium-on-a-chip to test the causal factors from the mother (including extracellular vesicles) that may impact development. We have also begun to test how HS impacts the health of the offspring. Collectively these data allow us to understand the health implications of peri-implantation HS exposure as well develop the tools necessary to test mitigation strategies to reduce the health impacts across life-course health of humans in a warming world.
